چکیده

Phagocytosis of zymosan particles by human peripheral blood polymorphonuclear leukocytes (PMN) results in the release of lysosomal enzymes into the extracellular medium (1) and enhanced glucose oxidation and lactate production (2) . The antibiotic, cytochalasin B (CB), inhibits phagocytosis by PMN (3, 4), presumably by interfering with the membrane-associated network of microfilaments (5) . CB is therefore useful in studies of the interrelationship between phagocytosis and metabolic changes in stimulated PMN. This study confirms earlier reports that CB inhibits phagocytosis by PMN (3, 4, 6) and carbohydrate metabolism in phagocytosing PMN (4, 6, 7) . The release of the lysosomal enzyme, ,3-glucuronidase, however, was moderately stimulated by CB under conditions which resulted in almost total inhibition of phagocytosis .

Thromboxane generation by human peripheral blood polymorphonuclear leukocytes

Human peripheral blood polymorphonuclear leukocytes were stimulated to generate thromboxane B2 in a time- and concentration-dependent fashion upon exposure to serum-treated zymosan particles. Conversion by stimulated PMN of [14C] arachidonic acid to [14C]thromboxane B2 was confirmed by thin-layer radiochromatography, radio-gas chromatography, and mass spectrometry. Generation of thromboxane B2 ...

Potassium and Amino Acid Transport in Human Leukocytes Exposed to Phagocytic Stimuli

Influxes of potassium and amino acids were measured in suspensions of human polymorphonuclear leukocytes (PMNs) under resting conditions and after various phagocytic stimuli. Both ouabain-sensitive (or pump) and ouabain-insensitive (or leak) influxes of K were determined. In 5 mM external K, mean total K influx was 0.69 nmol/10(6) cells x min, of which 52% was ouabain-sensitive. Ouabain binding...

Modification of Zymosan-induced Release of Lysosomal Enzymes from Human Polymorphonuclear Leukocytes by Cytochalasin B

During the process of phagocytosis, polymorphonuclear leukocytes (PMN) release lysosomal enzymes into the extracellular medium. When the antibiotic cytochalasin B (CB) is present in the incubation medium along with phagocytable particles, enhanced recovery of enzyme activities from the incubation medium has been observed. These findings have led to the interpretation that CB enhances lysosomal ...

Subcellular Localization of H202 Production in Human Neutrophils Stimulated With Particles and an Effect of Cytochalasin-B on the Cells

The ultrastructural localization of H202 production in suspended polymorphonuclear leukocytes (PMN) stimulated with particles was studied using CeCI3 technique. PMN stimulated with opsonized zymosan or polystylene latex with or without lgG were incubated in 0.1 M Tris-maleate buffer with 1 mM CeCI, and 10 mM aminotriazole. Cells were then fixed and embedded in a resin for electron microscopy. T...

Activated polymorphonuclear neutrophils disseminate anti-inflammatory microparticles by ectocytosis.

On activation, human neutrophils release microparticles, called ectosomes, directly from the cell surface membrane. Microparticles from platelets, endothelial cells, and monocytes were reported to support coagulation or to modulate vascular homeostasis by activating monocytes as well as endothelial cells. We find that neutrophil ectosomes have no proinflammatory activity on human macrophages as...
